How to determine dot percentage of a printed halftone?

There are densitometers to know it precisely. But what is the general rule or idea to determine the percentage of an already printed halftone.

We have been known to physically measure a dot using a 100 power microscope that has a scale built into the viewing lens. Then knowing what the dot size on plate is compared to the printed dot you determine its gain.
